RCTRs are produced in mice challenged with Escherichia coli exudate. FVB mice were challenged with 5 × 105 E. coli for 24 hours. Then, lavage was performed with 5 mL of phosphate-buffered saline+/+ and was incubated with 30 μmol/L of DHA. Methanol fractions were analyzed and quantified for all sulfido conjugates. MCTRs regulate selected inflammatory cytokines with human macrophages. Proteome Profiler cytokine array was performed using human macrophages (2 × 106 cells). RCTRs do not alter survival of human neutrophils. Human polymorphonuclear cells (PMNs) were incubated for either 24 or 48 hours in phosphate-buffered saline (PBS) with calcium and magnesium (pH = 7.2, 37°C, 5% CO2), in vehicle PBS with calcium and magnesium containing 0.1% ethanol, or with the addition of 10 nmol/L RCTR1, RCTR2, RCTR3, or RvD1. Cell viability was assessed using flow cytometry with surface staining for annexin V and propidium iodide (PI; V13242; Invitrogen). Annexin+, PI+ represents apoptotic PMN populations. There were no statistically significant differences in apoptotic populations between all groups (two-way analysis of variance with Tukey's multiple comparison test). Data are expressed as means ± SEM. n = 3 healthy volunteers.
